Disorders of lipoprotein metabolism include primary and secondary conditions that substantially increase or decrease specific circulating lipids (e.g., cholesterol or TGs) or lipoproteins (e.g., low density or high density lipoproteins, see below). The demonstration that cholesterol-lowering therapy significantly reduces the clinical complications of atherosclerotic cardiovascular disease (ASCVD) makes it important for clinicians to be familiar with the diagnosis and treatment of lipoprotein disorders. This chapter reviews normal lipoprotein physiology, the pathophysiology of disorders of lipoprotein metabolism, the effects of genetic and environmental factors on lipoprotein metabolism, and the clinical approaches to the diagnosis and management of lipoprotein disorders.
